    Impact of limited solvent capacity on metabolic rate, enzyme activities, and metabolite concentrations of S. cerevisiae glycolysis

    The cell's cytoplasm is crowded by its various molecular components, resulting in a limited solvent capacity for the allocation of new proteins, thus constraining various cellular processes such as metabolism. Here we study the impact of the limited solvent capacity constraint on the metabolic rate, enzyme activities, and metabolite concentrations using a computational model of Saccharomyces cerevisiae glycolysis as a case study. We show that given the limited solvent capacity constraint, the optimal enzyme activities and the metabolite concentrations necessary to achieve a maximum rate of glycolysis are in agreement with their experimentally measured values. Furthermore, the predicted maximum glycolytic rate determined by the solvent capacity constraint is close to that measured in vivo. These results indicate that the limited solvent capacity is a relevant constraint acting on S. cerevisiae at physiological growth conditions, and that a full kinetic model together with the limited solvent capacity constraint can be used to predict both metabolite concentrations and enzyme activities in vivo. © 2008 Vazquez et al

    3D-nanoprinted antiresonant hollow-core microgap waveguide: an on-chip platform for integrated photonic devices and sensors.

    Due to their unique capabilities, hollow-core waveguides are playing an increasingly important role, especially in meeting the growing demand for integrated and low-cost photonic devices and sensors. Here, we present the antiresonant hollow-core microgap waveguide as a platform for the on-chip investigation of light-gas interaction over centimeter-long distances. The design consists of hollow-core segments separated by gaps that allow external access to the core region, while samples with lengths up to 5 cm were realized on silicon chips through 3D-nanoprinting using two-photon absorption based direct laser writing. The agreement of mathematical models, numerical simulations and experiments illustrates the importance of the antiresonance effect in that context. Our study shows the modal loss, the effect of gap size and the spectral tuning potential, with highlights including extremely broadband transmission windows (>200 nm), very high contrast resonance (>60 dB), exceptionally high structural openness factor (18%) and spectral control by nanoprinting (control over dimensions with step sizes (i.e., increments) of 60 nm). The application potential was demonstrated in the context of laser scanning absorption spectroscopy of ammonia, showing diffusion speeds comparable to bulk diffusion and a low detection limit.     Objetivo: Reflexionar sobre las características de la entrevista fenomenológica como parte de una investigación fenomenológica en investigación en enfermería y presentar algunas recomendaciones prácticas para su realización.Método: Estudio teórico reflexivo realizado en los meses de octubre y noviembre de 2016 sobre el desarrollo de la entrevista fenomenológica en enfermería, basado en la literatura científica y análisis crítico de los autores.Resultados: Se presentan las características de la entrevista fenomenológica y se sugieren algunas recomendaciones prácticas para su aplicación en enfermería.Conclusiones: La entrevista constituye un instrumento valioso para el acceso al fenómeno estudiado en enfermería y desvelado del ser.Palabras clave: Investigación cualitativa.     Integrating fluctuations into distribution of resources in transportation networks

    We propose a resource distribution strategy to reduce the average travel time in a transportation network given a fixed generation rate. Suppose that there are essential resources to avoid congestion in the network as well as some extra resources. The strategy distributes the essential resources by the average loads on the vertices and integrates the fluctuations of the instantaneous loads into the distribution of the extra resources. The fluctuations are calculated with the assumption of unlimited resources, where the calculation is incorporated into the calculation of the average loads without adding to the time complexity. Simulation results show that the fluctuation-integrated strategy provides shorter average travel time than a previous distribution strategy while keeping similar robustness. The strategy is especially beneficial when the extra resources are scarce and the network is heterogeneous and lowly loaded.Comment: 14 pages, 4 figure

    Modelling spatio-temporal data with multiple seasonalities: the NO2 portuguese case

    This study aims at characterizing the spatial and temporal dynamics of spatio-temporal data sets, characterized by high resolution in the temporal dimension which are becoming the norm rather than the exception in many application areas, namely environmental modelling. In particular, air pollution data, such as NO2 concentration levels, often incorporate also multiple recurring patterns in time imposed by social habits, anthropogenic activities and meteorological conditions. A two-stage modelling approach is proposed which combined with a block bootstrap procedure correctly assesses uncertainty in parameters estimates and produces reliable confidence regions for the space-time phenomenon under study. The methodology provides a model that is satisfactory in terms of goodness of fit, interpretability, parsimony, prediction and forecasting capability and computational costs. The proposed framework is potentially useful for scenario drawing in many areas, including assessment of environmental impact and environmental policies, and in a myriad applications to other research fields.     Comprehensive Analysis of Market Conditions in the Foreign Exchange Market: Fluctuation Scaling and Variance-Covariance Matrix

    We investigate quotation and transaction activities in the foreign exchange market for every week during the period of June 2007 to December 2010. A scaling relationship between the mean values of number of quotations (or number of transactions) for various currency pairs and the corresponding standard deviations holds for a majority of the weeks. However, the scaling breaks in some time intervals, which is related to the emergence of market shocks. There is a monotonous relationship between values of scaling indices and global averages of currency pair cross-correlations when both quantities are observed for various window lengths Δt\Delta t.Comment: 13 pages, 10 figure

    Transcription of toll-like receptors 2, 3, 4 and 9, FoxP3 and Th17 cytokines in a susceptible experimental model of canine Leishmania infantum infection

    Canine leishmaniosis (CanL) due to Leishmania infantum is a chronic zoonotic systemic disease resulting from complex interactions between protozoa and the canine immune system. Toll-like receptors (TLRs) are essential components of the innate immune system and facilitate the early detection of many infections. However, the role of TLRs in CanL remains unknown and information describing TLR transcription during infection is extremely scarce. The aim of this research project was to investigate the impact of L. infantum infection on canine TLR transcription using a susceptible model. The objectives of this study were to evaluate transcription of TLRs 2, 3, 4 and 9 by means of quantitative reverse transcription polymerase chain reaction (qRT-PCR) in skin, spleen, lymph node and liver in the presence or absence of experimental L. infantum infection in Beagle dogs. These findings were compared with clinical and serological data, parasite densities in infected tissues and transcription of IL-17, IL-22 and FoxP3 in different tissues in non-infected dogs (n = 10), and at six months (n = 24) and 15 months (n = 7) post infection. Results revealed significant down regulation of transcription with disease progression in lymph node samples for TLR3, TLR4, TLR9, IL-17, IL-22 and FoxP3. In spleen samples, significant down regulation of transcription was seen in TLR4 and IL-22 when both infected groups were compared with controls. In liver samples, down regulation of transcription was evident with disease progression for IL-22. In the skin, upregulation was seen only for TLR9 and FoxP3 in the early stages of infection. Subtle changes or down regulation in TLR transcription, Th17 cytokines and FoxP3 are indicative of the silent establishment of infection that Leishmania is renowned for. These observations provide new insights about TLR transcription, Th17 cytokines and Foxp3 in the liver, spleen, lymph node and skin in CanL and highlight possible markers of disease susceptibility in this model

    Molecular characterization of beta-tubulin from Phakopsora pachyrhizi, the causal agent of Asian soybean rust

    β-tubulins are structural components of microtubules and the targets of benzimidazole fungicides used to control many diseases of agricultural importance. Intron polymorphisms in the intron-rich genes of these proteins have been used in phylogeographic investigations of phytopathogenic fungi. In this work, we sequenced 2764 nucleotides of the β-tubulin gene (Pp tubB) in samples of Phakopsora pachyrhizi collected from seven soybean fields in Brazil. Pp tubB contained an open reading frame of 1341 nucleotides, including nine exons and eight introns. Exon length varied from 14 to 880 nucleotides, whereas intron length varied from 76 to 102 nucleotides. The presence of only four polymorphic sites limited the usefulness of Pp tubB for phylogeographic studies in P. pachyrhizi. The gene structures of Pp tubB and orthologous β-tubulin genes of Melampsora lini and Uromyces viciae-fabae were highly conserved. The amino acid substitutions in β-tubulin proteins associated with the onset of benzimidazole resistance in model organisms, especially at His 6 , Glu 198 and Phe 200 , were absent from the predicted sequence of the P. pachyrhizi β-tubulin protein
